# Cleaning Validation Protocol Template

## When to Use

- When qualified personnel plan evidence that a cleaning process meets approved acceptance limits.
- When equipment, residues, sampling, and analytical methods need process-specific justification.

## When NOT to Use

- To infer acceptable residue limits or sampling adequacy from generic example values.
- Do not represent author self-checks as independent or authorized approval. See [Self-approval](../../../../../docs/principles/anti-patterns.md#self-approval).

## Pairs Well With

- [Equipment Qualification Protocol Template](../../../../../domains/delivery/industry-specializations/healthcare-pharmaceutical/validation/equipment_qualification_protocol_template.md).
- [Validation Master Plan Template](../../../../../domains/delivery/industry-specializations/healthcare-pharmaceutical/validation/validation_master_plan_template.md).

Selection context: Delivery domain; universal methodology; advanced complexity. Match the situations above to project phase, risk, team size, and industry using the [decision-engine context model](../../../../../meta/architecture-research/800-801-context-assessment-model.md).

## Applying this template

Treat filled examples, dates, numerical limits, sampling frequencies, and approval outcomes as illustrations. Before execution, replace them with the approved product/site requirements, scientific rationale, method references, responsible reviewer, and acceptance evidence. Record the applicable jurisdiction and current guidance edition; a completed worksheet alone does not demonstrate regulatory compliance.

| Requirement or limit | Applicable source / edition | Product-specific rationale | Approval / date | Execution evidence |

- Toxicity: [Highest toxicity]
- Physical characteristics: [Most difficult physical properties, e.g., viscosity, adherence]

#### 4.1.2 Worst-Case Equipment Conditions

- Maximum operating time: [Longest production run time]
- Minimum cleaning agent concentration: [Lowest effective concentration]
- Minimum cleaning time: [Shortest effective cleaning time]
- Longest dirty hold time: [Maximum time between production end and cleaning start]

### 4.2 Sampling Locations

A total of [X] sampling locations have been identified based on:
- Direct product contact surfaces
- Difficult-to-clean areas
- Flow path/product stream
- Representative sampling of total surface area

| Location ID | Description | Rationale | Sampling Method |
|-------------|-------------|-----------|----------------|
| L1 | [e.g., Interior vessel bottom] | [e.g., Potential pooling point] | [Swab/Rinse] |
| L2 | [e.g., Agitator shaft] | [e.g., Difficult geometry, product contact] | [Swab/Rinse] |
| L3 | [e.g., Discharge valve] | [e.g., Product flow restriction point] | [Swab/Rinse] |
| ... | ... | ... | ... |

**Sampling Locations Diagram:** [Reference to diagram in Appendix]

### 4.3 Acceptance Criteria

#### 4.3.1 Visual Inspection
- All accessible product-contact surfaces must be visibly clean when inspected under [specify lighting conditions, e.g., 500 lux minimum]
- No visible residue, film, or discoloration

#### 4.3.2 Chemical Residue Limits
Limits for active pharmaceutical ingredient (API) and cleaning agents are established based on:

**For API residue:**
- Maximum Allowable Carryover (MAC) = (Minimum Daily Dose of Previous Product × Safety Factor) / Maximum Daily Dose of Next Product
- Limit in terms of concentration = MAC / Minimum Batch Size of Next Product
- Limit per surface area = Limit in terms of concentration × Minimum Batch Size / Total Shared Surface Area

**Example calculation:**
- Safety factor: 0.001 (1/1000)
- Minimum daily dose of previous product (Product A): 10 mg
- Maximum daily dose of next product (Product B): 100 mg
- Minimum batch size of Product B: 100 kg
- Total shared surface area: 5 m²

MAC = (10 mg × 0.001) / 100 mg = 0.0001 mg/mg = 0.1 μg/mg
Limit in terms of concentration = 0.1 μg/mg × 1,000,000 mg/kg = 100,000 μg/kg
Limit per surface area = 100,000 μg/kg × 100 kg / 5 m² = 2,000,000 μg/m² = 2 mg/m² = 2 μg/cm²

**For cleaning agent residue:**
- Based on supplier recommendations, toxicological data, and regulatory guidelines
- Typically 10 ppm or lower

#### 4.3.3 Microbiological Limits (if applicable)
- Total aerobic microbial count (TAMC): ≤ 100 CFU/100cm²

- Absence of specified organisms (e.g., E. coli, Salmonella, Pseudomonas aeruginosa, Staphylococcus aureus)
